If we use Directory Replicator service with a SQL database we often obtain an access denied message and the user can’t access de database ¿Can anyone help me?

Copying DB files while SQL has them open will generally result in MDF/LDF files that are not usable. If you're using this primarily as a backup system, you should consider setting up a scheduled task in SQL to dump a backup file to disk. Then you can replicate this/these files. On the other hand, if you're trying to maintain a standby server, you may want to look into something like log shipping.
